Essay about acid fast stain - 402 Words.

Acid-Fast Staining Acid-fast stain is a differential stain. More so, it is a physical property of certain bacteria, particularly a bacteria’s resistance to decolorization by certain acids during staining procedures. Most bacteria are decolorized by the use of acid-alcohol.

The acid-fast stain is performed on samples to demonstrate the characteristic of acid fastness in certain bacteria. Acid fastness is a characteristic that is shared by just a few organisms, so staining to determine if organisms possess this trait is useful in microbial identification schemes.

In 1882 acid fast staining was developed by Paul Ehrlich and was later successfully modified by Ziehl And Neelsen. Ehrlich first discovered that Mycobacterium tuberculosis, which is the main causing agent of tuberculosis, retained the primary stain given even after washing it with an acid-alcohol mixture.Gram staining provides a rapid method for the initial identification of bacteria involved in infections. Similarly, the acid-fast staining procedure helps to specifically identify the organisms belonging to the class of bacteria called Mycobacteria, such as Mycobacterium tuberculosis.Technique. Acid fuchsin is a magenta red acid dye that is largely used for plasma staining whereas basic fuchsin is a magenta basic dye largely used to stain the nucleus. The technique is also referred to as acid fast staining. The acid fast bacteria have a waxy substance (mycolic acid) on their cell wall that makes them impermeable to staining procedures.

Acid-fast bacteria include the Mycobacteria and some of the Nocardia. The acid-fast staining property results from the presence of membrane glycolipids and very long chain 2-alkyl-3-hydroxy fatty acids (mycolic acids) bound to the peptidoglycan. In mycobacteria, up to 60% of the dry weight of the cell envelope consists of lipid.Acid fast cell walls are so durable that the stain (carbol fuschin) must be driven into the cells with heat. The cells are then decolorized with acid-alcohol, all other cells will decolorize with this strong solvent, but acid fast bacteria will not.

Experiment on performing acid-fast staining of a bacteria to find out whether it is acid-fast or non-acid-fast! Purpose: Most of the bacteria can be stained, either by simple basic staining or by gram staining, as their cells take up stains easily.

Principle of acid-fast staining: Acid-fastness has been attributed to the high content of lipids, fatty acids and higher alcohols, which constitute almost 40% of the dry weight of tubercle bacilli. Mycolic acid (a wax), acid-fast in the free state, is found in all acid-fast bacteria.

Gram positive bacteria (with a thicker peptidoglycan layer) retain crystal violet stain during the decolourisation process, while Gram negative bacteria lose the crystal violet stain and are instead stained by the safranin in the final staining process.6 From our observation from simple staining of Bacillus sp ,it can be seen that the bacteria is rod-shaped.

Most samples that are submitted for acid-fast bacilli (AFB) testing are collected because the healthcare practitioner suspects that a person has tuberculosis (TB), a lung infection caused by Mycobacterium tuberculosis. Mycobacteria are called acid-fast bacilli because they are a group of rod-shaped bacteria (bacilli) that can be seen under the microscope following a staining procedure where.

There are several types of staining through which the bacteria can be identified based on the color and shape. The staining methods used in the lab are Gram Staining, Capsule Staining, Endospore Staining, and Acid Fast staining. One of the most significant method of staining is the Gram Staining, as it is highly dependent (McCarthy, 25).
